Post-Doctoral Fellow in Cancer Biology: Philadelphia, PA, United States

The Amaravadi lab at the University of Pennsylvania works on understanding the role of autophagy as a resistance mechanism in cancer therapy. We are working on multiple studies recently funded by 5 year NIH grants including 2 RO1s, PO1 and SPORE.

Specific Projects that could be the responsibility of the successful applicant include:
1) Dissecting the molecular pathway that connects mutant BRAF and MAPK signlaing with the ER stress response and autophagy. This project will work closely with another PI that is an ER stress expert.

2) Determing the role of autophagy as a resistance mechanism to immunotherapy. This project will utilize
existing melanoma SPORE infrastructure to collect T cells and tumor cells from the same patient and establish a 3D Culture platform to study the impact of immune checkpoint inhibitors on tumor cells. Specifically we will use proteomics to determine the stress response pathways that contribute to resistance to immunotherapy. We will work closely with immunobiology experts.

3)Development of novel lysosomal autophagy inhibitors for cancer therapy including high throughput compound screening, functional validation, target identification and validation. This is a well established collaboration with chemistry and an industry partner.

4) Proteomics- based approach to novel predictive and pharmacodynamic biomarker discovery and validation in three dimensional culture systems, patient derived xenograft models and clinical patient specimens using BRAF mutant melanoma as a model system.

5) Development of a multigene prediction signature that identifies tumors that are sensitive to autophagy inhibitors.

The post-doctoral fellow will be paid a salary on an NIH equivalent scale commensurate with experience. This position will require interactions with basic scientists and clinicians. It will expose the fellow to team science approaches, clinical trials and translational studies, and pharmaceutical industry partnerships. The expectation would be generation of first author publications, independent grant funding, and laboratory management skills with close mentoring designed to develop an independent principal investigator. There is grant funding for 5 years for salary support.
Qualifications We are looking for a post-doctoral fellow with the following qualifications:

1) US citizen or permanent resident
2) Excellent English language writing and speaking skills
3) Publication history
4) Graduate work in cancer biology within the past 5 years
5) Secondary expertise in pharmacology, cell signaling, or immunology
6) Previous experience with xenograft or transgenic mouse models….
