PhD position (molecular biology): DNA demethylation and development: Mainz, Germany
In the field of “DNA demethylation and development” the research group of Prof. Christof Niehrs offers the following PhD project:
Developmental epigenetics in Xenopus embryos
Description:
In this project we will carry out a screen for novel epigenetic and demethylation regulators in Xenopus (frog) embryos, where microinjection yields facile access to functional analysis of gene function.
We will misexpress candidate epigenetic regulators by mRNA overexpression and antisense Morpholino injection to score for interesting developmental phenotypes. Candidate genes will then be studied by next generation sequencing, including genome-wide gene expression (RNA-seq) to monitor marker gene expression changes and DNA methylation (bisulfite-seq) analysis. Further characterization of changes in embryonic gene expression will be followed up by genome wide ChIP-seq and bioinformatics analysis. We will employ a systems biology approach for the understanding of the candidates gene function.
