Description:
The Division of Molecular Genome Analysis at the DKFZ aims at understanding molecular mechanisms leading to the transformation of the tumor microenvironment and its role in cancer initiation and tumor progression. A further hallmark of tumorigenesis is a reprogramming of cellular metabolism, and several oncogenes contribute to this process. Using an approach combining perturbation experiments and time-resolved proteomics and transcriptomics we aim to elucidate the impact of breast-cancer relevant oncogens on metabolic transformation.
Aim of this PhD project is the identification of proteins that can potentially serve as drug targets to revert the transformed metabolism of cancer cells. The successful candidate will use different breast cancer cell lines as model systems and monitor the impact of nutritional stress (high glucose, high load of fatty acids, insulin) and of inflammatory cues (e.g. cytokines). Identified targeted genes will be validated by targeted proteomics (reverse phase protein arrays) and by cellular assays. Quantitative proteomic data generated throughout this project will feed into a systems biology modeling approach that integrates metabolic pathways and cellular signaling.

We are looking for a highly motivated candidate with a vivid interest in cellular signaling, metabolism and breast cancer biology. Applicants should hold an above-average Diploma/Master's degree in biochemistry, biology or related fields. Experiences in basic methods of cellular biology, protein chemistry and molecular biology are a prerequisite.
