Plasma peptidomics for the identification of CRS biomarkers
The PdD student will identify diagnostic and predictive biomarkers for cardiorenal syndrome in plasma. Based on vast peptidomics experience of the IMCAR institute to identify disease mediators, PdD studen will generate high-throughput pep-tidomics profiles using LC-MS/MS on already available plasma samples for the cohorts (n~100/cohort; samples availalbe). PdD student will generate comprehensive lists of peptides associated with cardiorenal syndrome using a bioinformatics approaches. These will be used for molecular modelling of the cardiorenal syndrome by PdD student. After prioritization based on statistical significance and biological relevance, top biomarker candidates will be sequenced using MS and validated in independent samples (~50/patient cohort, samples already availalbe). Further, PdD student will be strongly involved in the identification of uremic toxins affecting the phenotype of VSMCs and cardiomyocytes by preparing chromatographic fractions from already ava!
ilable ur
emic dialysate for phenotypic screening assays, and by identifying the responsible toxins using mass-spectrometry.
