“Identification of the vascular function of the NADPHoxidase organizer Noxo1”
NADPH Oxidases of the Nox family are important sources of the cellular production of reactive oxygen species (ROS). The Nox-dependent ROS formation is controlled on multiple levels, among them interacting proteins. In epithelial cells, Noxo1 is an adapter protein which tethers Noxa1 to Nox1 and the plasma membrane and thereby maintains the Nox1-dependent NADPH oxidase active. Under certain conditions, Noxo1 expression increases also in the vascular system raising the possibility that this protein has an important impact on pathophysiological vascular ROS production.
The announced project particularly focuses on the protein Noxo1, which we observed to be expressed in the vascular system and for which we generated a knockout mouse. In the project, the expression control of Noxo1 in the vascular system will be studied and the physiological consequence of its loss in cultured cells and in the mouse will be determined in model system for the vascular system particularly focussing on angiogenesis and vascular regeneration in the context of diabetes.
Techniques included in the project concern methodology of cell and molecular biology, in particular for angiogenesis and as well as experiments of vascular regeneration in mice, determination of reactive oxygen species formation and analysis of animal and cell proteomics by LTQ-Orbitrap mass spectrometry particularly focussing on redox-modified proteins.
The Vascular Research Centre offers a well-equipped working environment where talented, ambitious, self-motivated students will find an ideal environment to develop and pursue their personal scientific ideas. The PhD student will become a member of the graduate schools of the collaborative research centre “Redox-Biology” funded by the German Research Foundation.
Candidates should hold a master degree in biology, biochemistry, pharmacology or a related field. Applicants coming from abroad are also encouraged to apply. While knowledge of the German language is not mandatory, excellent written and spoken English skills are required.
